Organometallic Flow Chemistry at Scale - A Highly Atroposelective Negishi Coupling Enables the Commercial Manufacturing Process of Divarasib

This webinar will focus on a novel atroposelective Negishi coupling process developed for large-scale manufacturing. Attendees will learn about the successful isolation of a late-stage advanced intermediate as a single isomer without the need for chromatography, as well as a continuous process that eliminates cryogenic conditions in the production of divarasib—a promising KRAS G12C inhibitor currently in Phase III clinical trials. The session will also explore the identification and characterization of the optimal catalyst, the role of additives in the reaction, the kinetics of the Negishi coupling, and the integration of metalation in flow chemistry.
